The Notch signalling pathway is an evolutionarily conserved pathway in multicellular organisms, which is vital for cellcell communication, important during fundamental developmental and physiological processes, including regulation of cell fate decisions during neuronal, cardiac and endocrine development, stem cell haematopoiesis, thymic Tcell development, and both tumor progression and suppression. Ligation of Notch receptors by their specific ligands, Jagged1(CD339), Jagged2, Delta like-1(DLL1), DLL3 and DLL4, on physically adjacent signal receiving cells, induces proteolysis of the receptors by ADAMfamily metalloproteases and gammasecretase complex, within the transmembrane domain, releasing the Notch intracellular domain (NICD) to translocate to the nucleus. Subsequent signal transduction then occurs through either the CSLNICDMastermind complex cascade (canonical pathway), or NFkappaBNICD and CSLNICD Deltex complex signalling cascades (noncanonical pathway). The canonical pathway inhibits the differentiation of stem cells or progenitor cells, whilst the noncanonical pathway promotes differentiation. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry and Functional assays. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ul labels 1x10e6 cells in 100ul Immunohistochemistry: Frozen and paraffin sections Optimal dilutions to be determined by the researcher. Hybridoma: P3U1 myeloma cells with spleen cells from Armenian hamsters. Recommended Positive Control: Mouse spleen Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.     Note: Applications are based on unconjugated antibody.
