- MaxLight™550 is a new Yellow-Green photostable dye conjugate comparable to Alexa Fluor™546, 555, DyLight™549 , Cy3™, TRITC and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (550nm); Emission (575nm); Extinction Coefficient 150,000. Applications: Suitable for use in FLISA, Western Blot, Immunoprecipitation, Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5–2ug/ml detects CFTR in RIPA lysates from CFTR-transfected BHK cells. Reported to detect CFTR from a human T84 colon carcinoma epithelial cell lysate. CFTR-transfected BHK cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with C9040-01 (0.5ug/ml). Proteins were visualized using IgG (HRP) goat anti-mouse secondary antibody and a chemiluminescence detection system. Note: Do not boil the lysate. Instead incubate at 37°C for 30 minutes. Immunoprecipitation: Immunoprecipitates CFTR from a T84 cell lysate. Immunocytochemistry: Reported to immunostain CFTR in T84 cells. Note: CFTR runs as a diffuse protein on SDS-PAGE. Optimal dilutions to be determined by the researcher. Recommended secondary antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™550 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
