Extra Details:: Applications: Suitable for use in ELISA, Western Blot, Immunoprecipitation, Immunocytochemistry. Other applications not tested. Recommended Dilution: Western Blot: 0.5–2ug/ml detects CFTR in RIPA lysates from CFTR-transfected BHK cells. Reported to detect CFTR from a human T84 colon carcinoma epithelial cell lysate. CFTR-transfected BHK cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with C9040-01 (0.5ug/ml). Proteins were visualized using IgG (HRP) goat anti-mouse secondary antibody and a chemiluminescence detection system. Note: Do not boil the lysate. Instead incubate at 37°C for 30 minutes. Immunoprecipitation: Immunoprecipitates CFTR from a T84 cell lysate. Immunocytochemistry: Reported to immunostain CFTR in T84 cells. Note: CFTR runs as a diffuse protein on SDS-PAGE. Optimal dilutions to be determined by the researcher. Recommended secondary antibodies: I1904-06C: IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo I1904-06H IgG, H&L (HRP) (X-Adsorbed) Pab Gt xMo Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
