- Cyclin D1 is a 36kD nuclear protein. The cell cycle in eukaryotic cells is mediated by the formation, activation, and deactivation of complexes containing Cyclin-Dependent Kinases (CDK) and cyclins. D-type cyclins usually associate with CDK4 (p34 PSK-J3). Complexes with CDK2 and CDK5 have also been reported (1). D-type cyclins are necessary for cells to achieve the G1 to S transition in the cell cycle (2). The D-type cyclins have been shown to associate with PCNA, the retinoblastoma gene product, p21, and other proteins. Applications: Suitable for use in ELISA, Western Blot, Immunoprecipitation, Immunohistochemistry. Other applications not tested. Recommended Dilution: ELISA: 0.1-1.0ug/ml Western Blot: 0.3-1.0ug/ml Immunoprecipitation: 5ug Immunohistochemistry: 2-10ug/ml. Formalin-fixed paraffin embedded sections require a Heat Induced epitope Retrieval (HIER) step prior to staining. Optimal dilutions to be determined by the researcher. Source: Ascites Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
