- Extra Details:
- Mouse C1rLP, also CLSPa, is a 55kD, secreted, atypical glycoprotein member of the AGY group of servine proteases. It has protease activity for pro-haptoglobin, but accomplishes cleavage without a typical Asp in the catalytic site. In addition, it is proposed to block unwanted complement-mediated proteolysis. Mature C1rLP is 443 amino acids (aa) in length. It contains one CUB domain (aa # 43-165), a truncated CCP2 region (aa # 167-223) and a serine protease domain (aa # 239-482). There is a potential short form that utilizes an alternate start site at Met 27. Over aa's # 44-456, mouse C1rLP is 78% and 90% aa identical to human and rat C1rLP, respectively. Applications: Western Blot: This antibody can be used at 0.1-0.2ug/ml with the appropriate secondary reagents to detect mouse C1rLP. The detection limit for rmC1rLP is approximately 2 ng/lane under non-reducing and reducing conditions. Immunoprecipitation: This antibody has been used to immunoprecipitate rmC1rLP from conditioned media of transfected NS0 cells. Direct ELISA: This antibody can be used at 0.5-1.0ug/ml with the appropriate secondary reagents to detect mouse C1rLP. The detection limit for rmC1rLP is approximately 0.07 ng/well. Optimal dilutions should be determined by each laboratory for each application. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
