- Collagens are highly conserved throughout evolution and are characterised by an uninterrupted "Glycine X Y" triplet repeat that is a necessary part of the triple helical structure. Type I collagen (95kD) is found in bone, cornea, skin and tendon. Mutations in the encoding gene are associated with osteogenesis imperfecta, Ehlers Danlos syndrome, and idiopathic osteoporosis. Reciprocal translocations between chromosomes 17 and 22, where this gene and the gene for Platelet-derived growth factor beta are located, are associated with a particular type of skin tumor called dermatofibrosarcoma protuberans, resulting from unregulated expression of the growth factor. Applications: Suitable for use in ELISA, Immunocytochemistry, Immunohistochemistry and Radioimmunoassay. Not recommended for Western Blots. Other applications not tested. Recommended Dilutions: Immunohistochemistry: 1:40-1:80 for immunofluorescent staining of frozen rat skin and liver tissues. The antibody is also reactive on paraffin embedded rat tissues (skin, liver) at a dilution of 1:500 using an ABC detection system. Immunocytochemistry: 1:40-1:80 using immunofluorescent staining of frozen rat skin and liver tissues. ELISA: 1:200 Optimal dilution determined by the researcher. Positive Control: Rat hepatic stellate cell lysate Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
