- PRAT4A (Protein associated with TLR4-A) is a ubiquitously expressed, 40kD protein that participates in Toll receptor expression. Mouse PRAT4A is 276aa in length. It contains a putative signal sequence of 37aa, followed by a 239aa mature region. Although the presence of a signal sequence suggests it is secreted, its best known activity involves an intracellular association with TLR4. PRAT4A appears to facilitate the maturation and expression of TLR4 on the cell surface. Studies suggest this involves the C-terminal 60aa. Mature mouse PRAT4A shares 90% and 96% aa identity with human and rat PRAT4A, respectively. Applications: Suitable for use in ELISA, Immunohistochemistry and Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 1ug/ml with Neuro‑2A mouse neuroblastoma cell line and RAW 264.7 mouse monocyte/macrophage cell line Immunohistochemistry: 1-15ug/ml in immersion fixed paraffin-embedded sections of mouse brain (thalamus) Optimal dilutions to be determined by the researcher. Storage and Stability: Lyophilized and reconstituted products are stable for 12 months after receipt at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
