- Clostridium botulinum neurotoxins are zinc-dependent proteases that block the release of acetylcholine from peripheral cholinergic nerve endings by targeting specific membrane proteins including synaptosomal-associated proteins (SNAP), vesicle-associated membrane proteins (VAMP), and syntaxins. Neurotoxin A is characteristically associated with human botulism and known to target and hydrolyse the 197-Gln--Arg-198 bond of SNAP-25. Applications: Suitable for use in ELISA, RIA, Immunofluorescence and Inhibition of BoTox-A binding to Neuro2A Cells. Other applications not tested. Recommended Dilutions: Immunofluorescence (IC): 1:50; stains Neuro2A cells pre-treated with GT1b ganglioside using indirect immunocytochemical staining procedure. Following treatment, cells were washed 4 times, fixed in a neutral buffered formalin for 10 minutes, permeablized and blocked. Reacted for 60 minutes with C5851-40B and then an IgG (FITC) goat anti-mouse. The staining pattern was observed using epifluorescent microscopy. Inhibition: Inhibits the binding of BoTox-A to Neuro2A cells treated with GT1b ganglioside prior to the addition of the toxin, and prior incubation of the antibody with the synthetic peptide. Tne immunogen has been shown to eliminate the inhibition of cellular binding by the antibody. Optimal dilutions to be determined by the researcher. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
