- MaxLight™ 490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm); Emission (515nm); Extinction Coefficient 73,000. Claudin-7 is one of the transmembrane proteins that seal tight junctions and is critical for maintaining cell-to-cell adhesion in epithelial cell sheets. Claudin-7 is localized in the intercellular junctions of the epithelial cells of the proximal tubules of the nephrons in mammalian kidney. It is reportedly down-regulated in head and neck squamous cell carcinomas and loss of claudin-7 expression has been linked to histological grade in ductal carcinoma and invasive ductal carcinoma. Claudin-7 has been shown to regulate the expression of prostate-specific antigen (PSA) and it is also expressed in normal human and mouse epithelia. Applications: Suitable for use in FLISA, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: FLISA: 0.1-1ug/ml Immunoprecipitation (Native): 2ug/IP reaction Western Blot: 1ug/ml Immunohistochemistry (FFPE): 5ug/ml Requires heat induced epitope retrieval (HIER) with EDTA, pH 8.0 prior to staining. Optimal dilutions to be determined by the researcher. Positive Control: Western Blot: T47D cells Immunohistochemistry: Human breast cancer tissues Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
