- Claudin-7 is one of the transmembrane proteins that seal tight junctions and is critical for maintaining cell-to-cell adhesion in epithelial cell sheets. Claudin-7 is localized in the intercellular junctions of the epithelial cells of the proximal tubules of the nephrons in mammalian kidney. It is reportedly down-regulated in head and neck squamous cell carcinomas and loss of claudin-7 expression has been linked to histological grade in ductal carcinoma and invasive ductal carcinoma. Claudin-7 has been shown to regulate the expression of prostate-specific antigen (PSA) and it is also expressed in normal human and mouse epithelia. Applications: Suitable for use in ELISA, Immunoprecipitation, Western Blot and Immunohistochemistry. Other applications have not been tested. Recommended Dilutions: ELISA: 0.1-1ug/ml Immunoprecipitation (Native): 2ug/IP reaction Western Blot: 1ug/ml Immunohistochemistry (FFPE): 5ug/ml Requires heat induced epitope retrieval (HIER) with EDTA, pH 8.0 prior to staining. Optimal dilutions to be determined by the researcher. Positive Control: Western Blot: T47D cells Immunohistochemistry: Human breast cancer tissues Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
