- The cyclin-dependent kinases form complexes with their cyclin partners and with cdk inhibitors. cdk6 and cdk4 associate with the D-type cyclins and target the retinoblastoma protein, their major substrate—a necessary event for entry into S-phase in cells not deficient for pRb (1). Heterotrimeric complexes of cdk6/ cyclin D are sequestered in the inactive form by their inhibitory component p27 Kip1 or one of the INK4 proteins (2). Dissociation and degradation of the inhibitory CKIs from the trimeric complex allow exit from the restriction point and progression through G1 (3). Unlike the INK/Kip CKIs, p21 Waf1/Cip1 enhances association of cdk4 and cdk6 with their cyclin D partners (4). Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:2000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
