- Human CD166 is a type 1 glycoprotein expressed on activated T cells, B cells and monocytes and appears to be the ligand for CD6 (1,2). Human CD166 may be important for activation of T cells. Applications: Suitable for use in Flow Cytometry and Antibody Blocking. Other applications not tested. Recommended Dilutions: Flow Cytometry: 1:50 (10ug/ml) Antibody Blocking: Blocks binding of CD6 to CD166. Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Human Raji cells were washed and incubated 45 minutes on ice with 80ul of C2548-90Mg at a dilution factor of 1:50 (10ug/ml). 2. Cells were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 1.29 log10 fluorescent units when compared to a Mouse IgG1/R-PE negative control at a similar concentration. 3. Binding was blocked when pre incubated 10 minutes with 20ul of 0.5mg/ml C2548-90M6 (anti-CD166 antibody). Storage and Stability: May be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Dilute only prior to immediate use. Stable for at least 12 months at 4°C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
