- Extra Details:
- Human CD152 (CTLA-4) is a cell surface glycoprotein expressed at low levels on activated T cells. CD152 is a high affinity receptor for the costimulatory molecules CD80 (B7-1) and CD86 (B7-2) and appears to function as a negative regulator of T cell activation. Functional Application: Blocks binding of CD152 (CTLA-4) Ig fusion protein to its CD80/CD86 receptor. Applications: Suitable for use in FLISA, Flow Cytometry, Immunohistochemistry and Immunocytochemistry. Other applications not tested. Recommended Dilutions: FACS/IC: 1:50 or use 1.6ul of concentrated antibody per 5x10e5 cells per test Immunohistochemistry: Frozen sections Optimal dilutions to be determined by the researcher. FACS Protocol: 1. Five x 10e5 CD152-muIg coated beads per tube were spun down and incubated for 45 minutes at 4°C with 80ul of C2548-83G2 at 1:50 (10ug/ml). 2. Beads were washed twice, fixed and analyzed using FACS. 3. Beads stained with a mean shift of 2.48 log10 fluorescent units when compared to a Mouse IgG1 (PE) isotype control. 4. Binding was blocked when beads were preincubated 10 minutes with 20ul of 0.5ug/ml C2548-83C. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
