- Extra Details:
- Human CD137 Ligand is a type II transmembrane protein constitutively expressed by monocytes, B cells neuroblastoma cells (1), and some activated T cells. The interaction of CD137 (4-1BB) and CD137L can supply a costimulatory signal to T cells through CD137. This costimulus seems to be more effective for CD8+ effector T cells than for CD4+ helper T cells (3). The interaction can also effect a reverse signal, inducing monocyte activation (2), or T cell inactivation and programmed death (4) through CD137L. Applications: Suitable for use in ELISA, Western Blot and Flow Cytometry. Other applications not tested. Recommended Dilution: Functional Application: Antibody does not block the interaction between CD137L and CD137. Optimal dilutions to be determined by the researcher. Flow Cytometry Procotol: 5x10e5 cultured Raji human tumor cells were incubated 45 minutes on ice with 80ul of T2040-17C at 10ug/ml. Cells were washed twice and incubated with IgG/FITC after which they were washed three times, fixed and analyzed by FACS. Cells stained positive with a mean shift of 0.29 log10 fluorescent units when compared to a Mouse IgG2a negative control at a similar concentration. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
