Extra Details:
Human CD134L is a type II membrane protein with homology to TNF which is expressed on activated B cells and activated endothelial cells. CD134L binds to CD134 present on activated T cells, providing a costimulatory signal. This pathway seems to be more effective for costimulation of CD4+ helper T cells than for CD8+ effector T cells. Blockade of this interaction in mouse abrogated immunological effects in several models of immflamation and rejection. Applications: Suitable for use in Flow Cytometry and ELISA. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ug/ml (1:50) labels 5x10e5 HUVEC cells Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Cultured human umbilical cord vein endothelial cells (HUVEC) were harvested by trypsinization. 2. Five x 10e5 cells per tube were washed and incubated for 45 minutes on ice with 80ul of C2515-40D at 10ug/ml (1:50). 3. Cells were washed three times, fixed and analyzed by FACS. 4. Cell stained positive with a mean shift of 0.89log10 fluorescent units when compared to a Mouse IgG1/R-PE at a similar concentration. 5. Binding was blocked when cells were pre incubated for 10 minutes with 20ul of C2515-40A (0.5mg/ml). Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap.
