- Extra Details:
- Human CD134L (OX-40 Ligand) is a type II membrane protein with homology to TNF which is expressed on activated B cells and activated endothelial cells. CD134L binds to CD134 present on activated T cells, providing a costimulatory signal. This pathway seems to be more effective for costimulation of CD4+ helper T cells than for CD8+ effector T cells. Blockade of this interaction in mouse abrogated immunological effects in several models of immflamation and rejection. Applications: Suitable for use in ELISA and Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ug/ml (80ul) Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Cultured HUVEC (Human umbilical cord vein endothethial cells) per tube were harvested by trypsinization. 2. After which they were incubated 45 minutes on ice with 80ul of C2515-40B at 10ug/ml. 3. Cells were washed twice and incubated with secondary reagent Streptavidin/R-PE (S7973-88P) after which they were washed three times, fixed and analyzed using FACS. 4. Cells stained positive with a mean shift of 1.04 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration. 5. Binding was blocked when pre-incubated with 20ul of 0.5mg/ml C2515-40A. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
