- Extra Details:
- Human CD122 (IL-2 Rb) associates with CD25 (IL-2Ra) to form a high affinity receptor for IL-2 and is expressed on activated T cells, B cells and monocytes. The cytoplasmic domain of CD122 is involved with signal transduction. Functional Application: Antibody inhibits binding of IL-2 to IL-2R beta (CD122) (1). Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 20ug/ml; 80ul labels 5x10e5 ficoll prepared human peripheral blood lymphocytes. Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Five x 10e5 Ficoll prepared human peripheral blood mononucear cells were stimulated by incubating 5 days with 5 mg/ml PHA-P (P3370-61). 2. After which they were incubated 45 minutes on ice with 80ul of C2460-01G at 20ug/ml. 3. Cells were washed twice and incubated with S7973-88P: Streptavidin (PE), after which they were washed three times, fixed and analyzed using FACS. 4. Cells stained positive with a mean shift of 0.38 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration. Binding was blocked when cells were pre incubated 10 minutes with 20ul of 0.5mg/ml of C2460-01F. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
