Extra Details:: Applications: Suitable for use in Immunohistochemistry, Flow Cytometry, Inhibition of Cell Attchment and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Immunohistochemistry: Acetone-fixed frozen human epithelial tissues including tongue, foreskin and ovary. Not for use on formalin-fixed tissue. Flow Cytometry: Stains human squamous cell carcinoma (SCC9), ovarian carcinoma (SKOV-3) and umbilical vein endothelium (HUVEC). Stains RKO or K562 cells transfected for CD104 expression. Does not stain monocytes, neutrophils, lymphocytes, T cells, B cells or platelets. Inhibition of Cell Attachment: Induces a sub-basal split at the basement membrane in non-wounded tissue. In addition, wound closure was significantly inhibited by ASC-8, as the epithelial tongue only covered 40% of the wound are 120 hours post wounding. These results demonstrate that the beta4 adhesion-blocking antidody may have adverse effects on normal tissue (Egles, C, et al., 2010) Immunoprecipitation: Precipitates PAGE bands of 66, 76, 120, 135 and 200kD (reduced) or 66, 110, 125, 130 and 190kD (non-reduced) from surface-biotinylated SCC9 cells. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
