- CD99 (MIC2, p30/32MIC2, Ewing’s Sarcoma Marker, E2 Antigen) is a transmembrane glycoprotein with a molecular weight of 30kD. It is expressed as a 28kD precusor that undergoes O-glycosylation. It consists of a single chain of 185 residues with a 100 amino acid extracellular region, an a helical transmembrane region, and a 35 amino acid intracellular region. The extracellular region has three EF hand motifs, a proline-rich region and a collagen-like region. Detection of CD99 in formalin-fixed, paraffin-embedded tissues is useful in differentiating Ewing’s sarcoma (ES) and peripheral/primitive neuroectodermal tumors (PNETs) from other small round cell tumors (SRCTs) that can develop during childhood. CD99 strongly stains the membrane and cytoplasm of cells in most cases of ES and PNET, whereas other SRCTs, such as neuroblastoma, rhabdomyosarcoma, and non-lymphoblastic lymphomas are usually negative. CD99 may also be useful in differentiating thymic epithelial tumors. Positive CD99 expression has been reported in a number of other tumors, including: lymphoblastic lymphoma, hemangiopericytoma, solitary fibrous tumor, synovial sarcoma, cutaneous neuroendocrine carcinoma, glioblastoma and astrocytoma, and others. CD99 staining has been reported in many normal cell and tissue types, including: pancreatic islet cells, Sertoli’s cells, granulosa cells, and cortical thymocytes. CD99 is expressed on the cell membrane of some lymphocytes, cortical thymocytes, and granulosa cells of the ovary. The antigen is also expressed by most pancreatic islet cells, Sertoli cells of the testis, and some endothelial cells. Mature granulocytes express very little or no CD99. MIC2 is strongly expressed on Ewing’s sarcoma cells and primitive peripheral neuroectodermal tumors. Applications: Suitable for use in Immunohistochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry (Formalin/paraffin): 1-2ug/ml for 30 min at RT (Staining of formalin/paraffin tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min.) Optimal dilutions to be determined by the researcher. Positive Control: Pancreas or Ewing’s sarcoma Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
