- CD94 cell surface antigen, also known as the Kp43 antigen, is expressed by NK cells and a subset of T lymphocytes. CD94 has been shown to exist in two forms, of molecular weight 39kD and 43kD. The expression of CD94 molecule appears restricted to natural killer (NK) cells and to a T lymphocyte subpopulation including a subset of gamma/+ T-cells, and some CD8+ CD56+ T-cells, mainly V gamma 2/V2. CD94 is found to associate with NKG2 to form a heterodimer which is involved in the inhibition of cell mediated cytotoxicity against cells bearing appropriate MHC class I allotypes. On NK cells the antigen structure apparently functions as a receptor for HLA-B7 on target cells. This interaction is inhibiting cytolytic activity of the NK cells and protects the cells carrying HLA-By. Initially selected for its capacity to inhibit IL2-dependent proliferation of NK-cells. Blocking the receptor on resting NK cells, triggers the Fc-dependent redirected lysis by these cells of HLA-B7 carrying target cells. Applications: Suitable for use in ELISA. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
