Extra Details:
CD94 cell surface antigen, also known as the Kp43 antigen, is expressed by NK cells and a subset of T lymphocytes. CD94 has been shown to exist in two forms, of molecular weight 39kD and 43kD. The expression of CD94 molecule appears restricted to natural killer (NK) cells and to a T lymphocyte subpopulation including a subset of gamma/+ T-cells, and some CD8+ CD56+ T-cells, mainly V gamma 2/V2. CD94 is found to associate with NKG2 to form a heterodimer which is involved in the inhibition of cell mediated cytotoxicity against cells bearing appropriate MHC class I allotypes. On NK cells the antigen structure apparently functions as a receptor for HLA-B7 on target cells. This interaction is inhibiting cytolytic activity of the NK cells and protects the cells carrying HLA-By. Initially selected for its capacity to inhibit IL2-dependent proliferation of NK-cells. Blocking the receptor on resting NK cells, triggers the Fc-dependent redirected lysis by these cells of HLA-B7 carrying target cells. Fusion Partners: Spleen cells from immunized BALB/c mice were fused with cells of the SP2/0 myeloma cell line. Applications: . Flow Cytometry: Use 10ul of the suggested working dilution to label 1 x 10e6 cells in 100ul. Recommended Secondary Reagents: F(ab')2 Rabbit anti mouse IgG: FITC F(ab')2 Rabbit anti mouse IgG: RPE Recommended Negative Controls: Mouse IgG1 Negative Control Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
