- Human CD94 expression is restricted to natural killer (NK) cells and a small subset of T lymphocytes. CD94 is a type II membrane glycoprotein of the c-type lectin superfamily and essentially lacks a cytoplasmic tail. Functional Application: C2442-02 inhibits IL-2 dependent proliferation of NK cells. Applications: Suitable for use in Flow Cytometry and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Immunohistochemistry: Frozen sections. Not recommended for paraffin. Flow Cytometry: 5ug/ml Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Ficoll prepared human peripheral blood mononuclear cells were preincubated ~10 min with 20ul of 300ug/ml human IgG (to reduce non specific binding) after which they were incubated 45 on ice with 80ul of C2442-02 at 5ug/ml. 2. Cells were washed twice and incubated with 2º reagent Gt xMo IgG (FITC), after which they were washed three times, fixed and analyzed using a FACS. A net 15% sub population of the cells stained positive with a mean shift of 1.27 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
