- Human CD71 is a type II transmembrane glycoprotein found on activated leukocytes. CD71 binds transferrin and mediates uptake of iron through endocytosis and recycling. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Protocol: -5x10e5 cultured HPB-MLT cells were washed and preincubated 5 minutes with 20 ml of 250 mg/ml human IgG (To block non specific binding). -Next they were incubated 45 minutes on ice with 80 ml of anti-CD71 antibody at 5 mg/ml. -Cells were washed twice and incubated with 2º reagent Goat anti-Mouse IgG/FITC, after which they were washed 3 times, fixed and analyzed using a BD FACS Calibur. -Cells stained positive (95% net) with a mean shift of 2.23 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration. Recommended Secondary Antibodies: I1904-06F: IgG, H&L (FITC) (X-Adsorbed) Pab Gt xMo Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
