- CD69, also known as very early activation (VEA) antigen, is a disulfide-linked transmembrane homodimer whose differentially glycosylated subunits range from 35-39 kD.1,2 It is a C-type lectin, most closely related to the NKR-P1 and Ly-49 NK cell-activation molecules.3 CD69 is widely expressed on hematopoietic cells, including lymphocytes, neutrophils and eosinophils.1-4 Although not detectable on resting lymphocytes, its expression is rapidly (within 2 hours) upregulated upon activation of T, B and NK cells, and neutrophils.4 Constitutive expression of CD69 on subsets of thymocytes suggests that it may be involved in regulation of developmental events in addition to its role in activation of a variety of hematopoietic cells.4,5 This antibody augments PMA-induced T-cell proliferation,1,6 and induces redirected lysis of Fc receptor-bearing target cells by NK cells.7 Applications: • Flow cytometry 1,2,4-7 • Immunohistochemistry (acetone-fixed, frozen sections) • Immunoprecipitation 1 • In vitro T cell and NK-cell activation 1,7 Recommended Dilution: Flow Cytometry: ≤ 1ug/10e6 cells Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20°C or colder. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
