Extra Details:
MaxLight™550 is a new Yellow-Green photostable dye conjugate comparable to Alexa Fluor™546, 555, DyLight™549 , Cy3™, TRITC and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (550nm); Emission (575nm); Extinction Coefficient 150,000. Macrosialin is a heavily glycosylated transmembrane protein of 87-115kD, which is specifically expressed by tissue macrophages, Langerhans cells and at low levels by dendritic cells. Macrosialin is the murine homologue of the human macrophage glycoprotein CD68, both are members of the lysosomal-associated membrane protein (lamp) family which are located predominantly within the cells and can be detected by flow cytometry using cell permeablization. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry, Immunoprecipitation, Immunofluorescence and Western Blot. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ul labels 10e6 cells in 100ul. Membrane permeabilization is required. Immunohistochemistry (Paraffin): Heat induced epitope retrieval is recommended prior to staining paraffin section. Sodium citrate buffer, pH 6.0 is recommended. Immunohistochemistry: Frozen sections Western Blot: Non-reducing conditions recommended. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™550 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
