- Human CD58 is a cell surface glycoprotein that functions as an adhesion molocule and mediates costimulatory activity through its ligand CD2, which is expressed on T and natural killer cells (1,2). CD58 has wide cellular distribution on both hemopoietic and non-hemopoietic cells with strong expression on macrophages and memory T cells. Functional Application: Antibody will inhibit HLA-DR mediated T cell cytotoxicity. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Immunohistochemistry: Frozen. Flow Cytometry: 5x10e5 cultured human Nalm-6 cells were washed and incubated 45 minutes on ice with 80ml of anti-CD58/Biotin at 10mg/ml. Cells were washed twice and incubated with 2° reagent Streptavidin/R-PE after which they were washed three times, fixed and analyzed using a BD FACS Calibur. Cells stained positive with a mean shift of 1.29 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration. Binding was 94% blocked when pre incubated 10 minutes with 20ml of 0.5mg/ml anti-CD58 antibody. Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
