Extra Details:
Human CD49b is an integrin alpha 2 subunit that forms a heterodimer with beta 1 integrin and functions as an adhesion molecule. Applications: Suitable for use in Flow Cytometry and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 5ug/ml (80ul) labels 5x10e5 cultured Squamous cell carcinoma. Optimal dilutions to be determined by the researcher. FACS Protocol: 1. Five x 10e5 cultured UM-SCC (squamous cell carcinoma) cells were harvested by trypsinization, were washed and preincubated , -5’- -3’minutes will 20ul of 250ug/ml human IgG( to block nonspecific binding). After cells were incubated for 45 minutes on ice with 80ul of C2404-07K at a 1:50 dilution (5ug/ml). 2. Cells were washed three times, fixed and analyzed by FACS. 3. Cells stained positive with a mean shift of 0.92 log10 fluorescent units when compared to a Mouse IgG2a/FITC negative control at a similar concentration. 4. Binding was blocked when pre-incubated for 10 minutes with 20ul of 0.5mg/ml of C2404-07H. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
