Leukocyte common antigen is expressed exclusively on the surface of hematolymphoid cells and their progenitors, including T and B lymphocytes, granulocytes, monocytes, and macrophages. Mature erythrocytes and megakaryocytes do not express CD45. The CD45 antibody is an important component of the antibody panel used to distinguish hematolymphoid neoplasms from non-hematolymphoid neoplasms, including undifferentiated or anaplastic large cell tumors (malignant lymphoma, melanoma, and carcinoma). In a study of nasophryngeal biopsies, LCA staining was positive for all malignant melanomas and negative for all nasopharyngeal carcinomas. LCA may be used together with cytokeratin antibodies to identify the cell-lineage of anaplastic tumors, demonstrating the ability to identify lymphoma and carcinoma in up to 88% of cases in a single study. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: Neat; 10ul labels 10e6 cells in 100ul Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
