Extra Details:
CD45 or Leukocyte Common Antigen (LCA/T200) family is comprised of at least four isoforms (CD45RA, CD45RB, CD45RC, and CD45RO) of membrane glycoproteins with MW of 180-220kD. It consists of a large, heavily glycosylated, amino-terminal extracellular domain of 400-550 amino acids, a single transmembrane region and a large carboxy-terminal cytoplasmic domain of 700 amino acids. CD45/LCA is expressed on hematopoietic cell lines but absent on non-hematopoietic cell lines, normal and malignant non-hematopoietic tissues. The intracellular portion of CD45 proteins has phosphatase activity and is involved in regulation of transmembrane signals, functioning as a leukocyte membrane receptor capable of transducing signals via its phosphotyrosine phosphatase domains.Several studies on the function of CD45 in leukocytes support a crucial role of CD45 in the regulation of leukocyte activation, development, and function. In these processes, distinct CD45 isoforms appear to play differential functions. The diversity of the structure, size, and carbohydrate content of different CD45 isoforms is cell-type dependent and developmentally regulated. This structural heterogeneity is mainly the result of the alternative splicing of a single CD45 gene. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: Neat-1:5; 10ul labels 10e6 cells in 100ul Optimal dilutions to be determined by the researcher. Hybridoma: Spleen cells from DA rats were fused with cells of the Y3/A1.2.3 rat myeloma cell line Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
