Extra Details:
CD45 or Leukocyte Common Antigen (LCA/T200) family is comprised of at least four isoforms (CD45RA, CD45RB, CD45RC, and CD45RO) of membrane glycoproteins with MW of 180-220kD. It consists of a large, heavily glycosylated, amino-terminal extracellular domain of 400-550 amino acids, a single transmembrane region and a large carboxy-terminal cytoplasmic domain of 700 amino acids. CD45/LCA is expressed on hematopoietic cell lines but absent on non-hematopoietic cell lines, normal and malignant non-hematopoietic tissues. The intracellular portion of CD45 proteins has phosphatase activity and is involved in regulation of transmembrane signals, functioning as a leukocyte membrane receptor capable of transducing signals via its phosphotyrosine phosphatase domains. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry and Immunoprecipitation. Other applications have not been tested. Recommended Dilution: Immunohistochemistry: Frozen sections Immunohistochemistry (Paraffin): Requires antigen retrieval using heat treatment prior to staining of paraffin sections. EDTA, pH8.0 is recommmeded. Optimal dilutions to be determined by the researcher. Hybridoma: NS1 myeloma cells with spleen cells from Balb/c mice. Recommended Positive Control: Human Tonsil Recommended Negative Control: I1904-83P1: IgG2a Murine Negative Control Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.  Note: Applications are based on unconjugated antibody.
