- Extra Details:
- Human CD45 is expressed on all cells of hematopoietic origin, except erythrocytes. CD45 is a transmembrane tyrosine phosphatase which can exist in at least nine different isoforms resulting from tissue-specific alternative RNA splicing of exons 4-7 of a single gene coding for the various N-terminal peptide segments. CD45 plays an essential role in lymphocyte activation. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50 (or use 1.6ul of concentrated stock per 5x10e5 cell test). Optimal dilutions to be determined by the researcher. FACS Protocol: 1. Five x 10e5 cultured human peripheral blood mononuclear cells were washed and and preincubated ~10 minutes with 20ul human IgG at 250 ug/ml (to block nonspecific binding), after which they were incubated for 45 minutes on ice with 80ul of C2399-02F at a 1:50 dilution (10ug/ml). 2. Cells were washed three times, fixed and analyzed by FACS. 3. Cells stained positive with a mean shift of 2.4 log10 fluorescent units when compared to a IgG1 Murine Negative Control (PE) at a similar concentration. 4. Binding was blocked when pre-incubated for 10 minutes with 20ul of C2399-02C nat 0.5mg/ml Storage and Stability: May be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Dilute only prior to immediate use. Stable for 12 months at 4°C. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light.
