- Human CD35 binds complement components C3b, C4b, and iC3b and is expressed on erythrocytes, B cells, monocytes, granulocytes and folicular dendritic cells and on subsets of thymocytes. CD35 on erythrocytes mediates clearance of immunue complexes. Applications: Suitable for use in ELISA. Western Blot and Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ug/ml labels 5x10e5 cultured U-937 human tumor cells Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. 5x10e5 cultured U-937 human tumor cells were washed and pre-incubated 5 minutes with 20ul of 250ug/ml human IgG (to block non specific binding) after which they were incubated 45 min on ice with 80ul of C2387-04F at 10ug/ml. 2.Cells were washed twice and incubated with IgG (FITC) goat anti-mouse, after which they were washed three times, fixed and analyzed by FACS. 3.Cells stained positive with a mean shift of 0.70 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
