- CD31 (PECAM-1) is a member of the immunoglobulin superfamily. This heavily glycosylated protein is found in the entire vascular endothelium of adult mice and functions in mediating cellular adhesion by heterophilic and homophilic mechanisms. CD31 is detected within the lymphopoietic islands in the spleen of newborn (day 12) and in the bone marrow of adult mice. Capillary endothelial cells of adult mice also express CD31. Applications: Suitable for use in Immunofluorescence, Flow Cytometry, Immunoprecipitation and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: Stains the extracellular domain of mouse CD31. 10e6 cells were used per sample. No fixation or permeabilization needed. Epitope is sensitive to trypsin. Immunoprecipitation: Lysate of 35S-Methionine labeled 5HV cells was used resulting in a molecule of ~130kD. Immunohistochemistry (Frozen): 1:50. Tissue sections were fixed in acetone for 10 min at RT and pretreated with 0.01% hydrogen peroxide to quench endogenous peroxidases. Normal rabbit serum was used as blocking agent. Optimal dilutions to be determined by the researcher. Positive Control: Mouse t-end.1 cells Negative Control: L929 cells (fibroblasts) Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
