- Human CD28 is an important costimulatory molecule found on all CD4+ T cells and on about half of the CD8+ T cells. T cell activities attributed to CD28 include prevention of anergy, induction of cytokine gene transcription, stabilization of cytokine mRNA and activation of CD8+ cytotoxic T lymphocytes. Anti-CD28 antibodies have recently been shown to have a strong adjuvant effect when conjugated to the immunogen (4). The ligands for CD28 identified as CD80 (B7-1) and CD86 (B7-2), are immunglobulin superfamily monomeric transmembrane glycoproteins of 60kD and 80kD respectively. Applications: Suitable for use in ELISA and Flow Cytometry. Other applications not tested. Recommended Dilution: Immunohistochemistry: Frozen sections Flow Cytometry: 5ug/ml labels 5x10e5 cells Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Five x 10e5 cultured HPB-MLT cells per tube were washed and incubated 45 minutes on ice with 80ul of C2380-03A at 5ug/ml. 2.Cells were washed twice and incubated with Goat anti-Mouse IgG/FITC after which they were washed three times, fixed and analyzed by FACS. 3. Cells stained positive with a mean shift of 1.26 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
