- Human CD26 is a T cell activation antigen which can be associated with costimulation. The significance of the dipeptidyl peptidase IV activity remains unknown. Recent characterization of adenosine deaminase binding to CD26 suggests an important immunoregulatory role. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ug/ml Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Ficoll prepared human peripheral blood monouclear cells were stimulated in culture 1 day with 10ng/ml PMA and 1uM Ionomicin. 2. 5x10e5 cells per tube were washed and preincubated for ~5 minutes with 20ul of human IgG at 250ug/ml (to block non specific binding). 3. Cells weere then Incubated for 45 minutes on ice with 80ul of C2277-06D at 10ug/ml. 4. Cells were washed twice and incubated with secondary reagent, IgG (FITC), GtxMo 5. Washed three times, fixed and analyzed using FACS. Cells stained positive with a mean shift of 0.31 log10 fluorescent units when compared to a mouse IgG2b Negative control at a similar concentration. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
