- Extra Details:
- CD23 is a low affinity receptor for IgE (FceR) and both the FceRIIa and FceRIIb are expressed on mature IgD+/IgM+ B cells. The FceRIIb form is also present on monocytes, activated macrophages, eosinophils, platelets and follicular dendritic cells. CD23 is also present in a soluble form in serum. CD23 together with CD19 and CD22, plays a role in the modulation of B cell activation through membrane immunoglobulin and is involved in regulation of IgE synthesis. Expression of CD23 is increased by IL-4 and down-regulated by B-cell growth factor (BCGF). Binding of IgE immune complexes to CD23 increases the efficiency of B cell antigen processing and presentation. Via its interaction with CD21 it may play a role in adhesion processes. It mediates IgE-dependent cytotoxicity and phagocytosis by macrophages and eosinophils. CD23 is a 45kD glycoprotein, which is present on a subpopulation of freshly isolated peripheral blood and tonsil B cells and strongly expressed on EBV-transformed B lymphoblasts. The CD23 molecule is identical to the low affinity IgE receptor found on B cells. Expression of CD23 has been detected in neoplastic cells from cases of B cell chronic lymphocyctic leukemia and some cases of centroblastic/centrocytic lymphoma. The CD23 clustered antibodies give a characteristic binding pattern with follicular dendritic cells but the antigen is absent on plasma cells and B cell progenitors. Expression of the CD23 antigen is increased as B lymphocytes pass from a resting to an active phase. Cleavage of the membrane bound form releases a series of soluble fragments all of which still bind IgG except for the 12kD moiety. Recommended Negative Controls: Mouse IgG2a Negative Control: FITC Flow Cytometry: Use 10ul of the suggested working dilution to label 10e6 cells in 100ul. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: FITC conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
