- Extra Details:
- Antibodies to human CD17 react with lactosylceramide which is expressed on granulocytes, monocytes, platelets, and basophils (1). CD17 expression is downmodulated on activated granulocytes. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 10ug/ml. 80ul labels 5x10e5 cultured Nalm-6 human tumor cells. Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Wash and preincubate 5x10e5 cultured Nalm-6 human tumor cells for 5 minutes with 20ul of 250ug/ml human IgG to block non specific binding. 2. Incubate the cells with 80ul of C2268-50B at 10ug/ml for 45 minutes on ice. 3. Wash cells twice. 4. Incubate with Streptavidin (PE). 5. Wash cells three times, fix and analyze by FACS. Cells stained positive with a mean shift of 2.13 log10 fluorescent units when compared to a Mouse IgM/Biotin negative control at a similar concentration. Binding was blocked when pre-incubated 10 minutes with 20ul of 0.5mg/ml of C2268-50. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
