- Antibodies to human CD17 react with lactosylceramide which is expressed on granulocytes, monocytes, platelets, and basophils (1). CD17 expression is downmodulated on activated granulocytes. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Five x10e5 cultured Nalm-6 human tumor cells were washed and preincubated 5 minutes with 20ul of 250ug/ml human IgG (To block no specific binding) after which they were incubated 45 minutes on ice with 80ul of C2268-50 at 10ug/ml. 2. Cells were washed twice and incubated with IgM (FITC) Goat anti-Mouse after which they were washed three times, fixed and analyzed by FACS. 3. A Cells stained positive with a mean shift of 1.1 log10 fluorescent units when compared to a Mouse IgM negative control at a similar concentration. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
