Extra Details:
MaxLight™650 is a new Far-IR stable dye conjugate comparable to Alexa Fluor™647, DyLight™649, Cy5™ and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (655nm); Emission (676nm); Extinction Coefficient 250,000. The CD14 molecule is found predominantly on monocytes and macrophages in Flow Cytometry; it is less strongly expressed on granulocytes, and is absent from stem cells and myeloid cells of very early differentiation states. In immunohistochemistry, the CD14 molecule is found to be present on Langerhans cells, follicular dendritic cells, histocytes and high endothelial venules. In FLISA, recognizes the soluble form CD14. Reported to block the binding of bacterial lipopolysaccharide (LPS) to monocytes (2). Removal of sodium azide is recommended prior to use in functional assays. Applications: Suitable for use in Western Blot, Immunoprecipitation, FLISA and Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: Neat Use 10ul to label 10e6 cells in 100ul. Western Blot: Non-reducing conditions Optimal working dilutions to be determined by researcher. Hybridoma: Spleen cells from mice immunized BALB/c mice were fused with cells from the NS1-Ag4/1 mouse myeloma line. Recommended Negative Controls: Mouse IgG1 Negative Control Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add glycerol (40-50%) and aliquot. Freeze at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Note: Applications are based on unconjugated antibody.
