- Human CD9 is strongly expressed on platelets, on early B cells and on activated T cells. CD9 is a member of the tetraspan family crossing the membrane four times. Both the N-terminal and C-terminal regions are on the cytoplasmic side of the plasma membrane. Recent studies indicate CD9 may be involved with calcium mobilization. Applications: Suitable for use in Flow Cytometry. Other applications not tested. Recommended Dilution: Flow Cytometry: 1:50 Optimal dilutions to be determined by the researcher. FACS Protocol: 1. Cultured Nalm-6 human tumor cells were washed two times. 2. Five x 10e5 cells per tube were incubated 45 minutes on ice with 80ul of C2260-06E at 1:50 dilution (10ug/ml). 3. Washed three times, fixed and analyzed by FACS. Cells stained with a mean shift of 2.64 log10 fluorescent units when compared to a Mouse IgG1/PE negative control at a similar concentration. Binding was 99% blocked when pre-incubated for 10 minutes with 20ul of 0.5mg/ml of C2260-06B. A565/A280 Ratio: 3.16 Storage and Stability: May be stored at 4°C before opening. DO NOT FREEZE! Stable at 4°C as an undiluted liquid. Dilute only pror to immediate use. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity.
