- Extra Details:
- The most abundant CD8 antigen is CD8a/b heterodimer expressed mainly on 13-48% (about one-third of peripheral T cells) of peripheral blood lymphocytes–suppressor/cytotoxic T lymphocytes (Ts/Tc) and 70-80% of thymocytes. In addition, a proportion of gdT cells and NK cells express CD8aa homodimers. CD8a can form homodimers, but CD8b cannot. CD8b requires the presence of CD8a to be expressed on the cell surface. CD8 antigen is coreceptor for HLA class I molecule. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry and in vitro Blocking Studies. Other applications have not been tested. Recommended Dilutions: Flow Cytometry: Neat 1:10. 10ul labes 10e6 cells in 100ul. Immunohistochemistry: Reported to react with paraffin sections following PLP (periodate-lysine paraformaldehyde) fixation. Optimal dilutions to be determined by the researcher. Flow Cytometry Positive Control: Rat splenocytes Recommended Negative Controls: I1904-79C: IgG1 Murine Negative Control (PE) Hybridoma: NS1 myeloma cells with spleen cells from Balb/c mice. Storage and Stability: Lyophilized powder may be stored at 4°C. Reconstitute by adding sterile ddH2O. Aliquot and store at 4°C. DO NOT FREEZE! Reconstituted product is stable for 12 months as an undiluted liquid. Dilute only prior to immediate use. Freezing R-Phycoerythrin (PE) conjugates will result in a substantial loss of activity. PE conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
