Extra Details:
The human CD3/T cell receptor (TcR) complex is made up of at least five CD3 proteins (g, d, e, h, z) in association with either a/b or g/d proteins of the TcR. The TcR recognizes antigens in association with MHC molecules after which protein chains of the CD3 complex mediate activation signals triggered by TcR antigen binding. CD3 is expressed on greater than 95% of circulating human peripheral T cells. Applications: Suitable for use in Flow Cytometry, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 5ug/ml; 80ul labels 5x10e5 cultured human PBMC cells Immunohistochemistry: Paraffin sections Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Five x 10e5 prepared human PBMC cells were washed and incubated 45 minutes on ice with 80ul of C2254-01B at a concentration of 5ug/ml. 2. Cells were washed twice and incubated with S7973-88P: Streptavidin (PE), after which they were washed three times, fixed and analyzed by FACS using a lymphoid gate. 3. A 81% subpopulation of the cells stained positive with a mean shift of 2.16 log10 fluorescent units when compared to a Mouse IgG1/Biotin negative control at a similar concentration. 4. Binding was blocked when pre incubated 10 minutes with C2254-01A: CD3 (Early T Cell Marker). Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
