- The human CD3/T cell receptor (TcR) complex is made up of at least five CD3 proteins (g, d, e, h, z) in association with either a/b or g/d proteins of the TcR. The TcR recognizes antigens in association with MHC molecules after which protein chains of the CD3 complex mediate activation signals triggered by TcR antigen binding. CD3 is expressed on greater than 95% of circulating human peripheral T cells. Applications: Suitable for use in Flow Cytometry, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilution: Flow Cytometry: 5ug/ml Immunohistochemistry: Paraffin Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. Five x 10e5 human HPB-MLT cultured tumor cells were washed and and incubated 45 minutes on ice with 80ul of C2254-01AX at 5ug/ml. 2. Cells were washed twice and incubated with 50ul of IgG (FITC) Goat anti-Mouse. 3. After which they were washed three times, fixed and analyzed using a BD FACstar plus. 4. A net 99% population of the cells stained positive with a mean shift of 1.66 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
