- The human CD3/T cell receptor (TcR) complex is made up of at least five CD3 proteins (gamma, delta, epsilon, h, z) in association with either alpha/beta or gamma/delta proteins of the TcR. The TcR recognizes antigens in association with MHC molecules after which protein chains of the CD3 complex mediate activation signals triggered by TcR antigen binding. CD3 is expressed on greater than 95% of circulating human peripheral T cells. Applications: Suitable for use in Flow Cytometry, Western Blot and Immunohistochemistry. Other applications not tested. Recommended Dilutions: Flow Cytometry: 5ug/ml; 80ul labels 5x10e5 cells. Immunohistochemistry: Paraffin sections Optimal dilutions to be determined by the researcher. Flow Cytometry Protocol: 1. 5x10e5 ficoll prepared human PBMC were washed and incubated 45 minutes on ice with 80ul of C2254-01A at 5ug/ml. 2. Cells were washed twice and incubated with IgG (FITC) Gt xMo* 3. The cells were then washed three times, fixed and analyzed by FACS using a lymphocyte gate. 4. An 83% subpopulation of cells stained positive for CD3 with a mean shift of 1.99 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration. *Recommended Secondary Antibodies: I1904-23: IgG, X-Adsorbed (FITC) Pab Gt xMo I1904-04A: IgG, H&L (FITC) Pab Gt xMo I1904-06F: IgG, H&L (FITC) (X-Adsorbed) Pab Gt xMo I1904-14A: IgG, (FITC) Pab Gt xMo Storage and Stability: May be stored at 4°C for short-term only.
