The CD3 complex is closely associated at the lymphocyte cell surface with the T cell antigen receptor (TCR). Reportedly, CD3 complex is involved in signal transduction to the T cell interior following antigen recognition. The CD3 antigen is first detectable in early thymocytes and probably represents one of the earliest signs of commitment to the T cell lineage. In cortical thymocytes, CD3 is predominantly intracytoplasmic; however, in medullary thymocytes, it appears on the T cell surface. CD3 antigen is a highly specific marker for T cells and is present in majority of T cell neoplasms. CD3 antigen is expressed on approximately 60–80% of human peripheral blood lymphocytes, 20–40% of splenic lymphocytes, the majority of T-CLL and approximately 70% pf T-All. The CD3 complex consists of five different polypeptide chains (designated as gamma, delta, epsilon, zeta, and eta) with MW ranging from 16-28kD. Applications: Suitable for use in Flow Cytometry, Immunohistochemistry, Immunofluorescence and Western Blot. Other applications not tested. Recommended Dilutions: Flow Cytometry: 1:50-1:100 Requires membrane permeabilization Immunohistochemistry (Frozen): 1:100 Immunohistochemistry (Paraffin): 1:100. Requires antigen retrieval using heat treatment using Tris/EDTA buffer pH 9.0 prior to staining of paraffin sections. Optimal dilutions to be determined by the researcher. Positive for Immunohistochemistry: Tonsil Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
