- Caspase-7, also known as mammalian CED-3 homologue 3 alpha, Mch3 alpha, LAP3, or CMH1, is a 34kD protein which belongs to a unique family of CED/ICE cysteine proteases involved in cytokine activation and in the execution of apoptosis. Caspases are synthesized as precursors (pro-caspases) and they are converted into mature enzymes by apoptotic signals. The effects of caspases in apoptosis are accomplished by the cleavage of numerous proteins located in different intracellular compartments. Studies have demonstrated that at least three pro-caspases (pro-caspases-2,-3, and ñ9 were present in the mitochondrial as well as cytosolic fractions of untreated Jurkat T lymphocytes and only pro-caspase-2 was found in the nuclear fraction. Pro-caspases-7 and ñ8 were found only in the cytosolic fraction. In apoptotic cells, caspases-3,-8 and ñ9 were present in the cytosolic fraction whereas caspases-3 and-9 were also found in the mitochondrial fraction and caspase-7 in the microsomal fraction. Caspases-2 and-3 were present in the nuclear fraction. The selective localization of pro-caspases in different subcellular compartments may play an important but yet unknown role in their activation. These results indicate that translocation of active caspases to other subcellular compartments appears to be critical for the development of the apoptotic process (1). Caspases can be classified into two subclasses according to the length of their N-terminal prodomains. Caspases-1,-2,-8,-9 and-10 contain long prodomains and they are believed to act as upstream caspases which are activated through interaction with specific adapter proteins. Long prodomain caspases such as caspase-8 and caspase-9 are believed to act mainly as upstream caspases to cleave downstream short prodomain caspases such as caspases-3,-6 and-7 (2,3). Caspases-3,-6 and-7 have been implicated in the execution phase of apoptosis and are shown to cleave specific substrates as the cell begins to present the characteristic morphological changes such as nuclear condensation, cell blebbing and formation of apoptotic bodies. Caspase-3 was shown to cleave a wide range of cytoplasmic and nuclear proteins suggesting an important role for this protease in apoptosis (4). Caspase-7 is a close homolog of caspase-3 and these two caspases possess some quarternary structural characteristics that support their unique role among the caspase family (5). Studies with caspase-3 knock-out mice suggest that caspase-7 has distinct but possibly overlapping roles in apoptosis (6). Applications: Suitable for use in Western Blot and Immunohistochemistry.
