- MaxLight™490 is a new Blue-Green photostable dye conjugate comparable to DyLight™488, Alexa Fluor™488 and offers better labeling efficiency, brighter imaging and increased immunodetection. Absorbance (491nm); Emission (515nm); Extinction Coefficient 73,000. The catenins are polypeptides that bind to the conserved cytoplasmic tail of cadherins and are required for cadherin function. a-catenin is a 102kD protein that is associated with the cadherin indirectly through its interaction with b-catenin and g-catenin. b-Catenin and its homolog plakoglobin associate directly with the cadherin. Thus, the cell-cell adherens junction is a structure composed of the transmembrane cadherin, which is associated directly with either b-catenin or plakoglobin, which in turn assoctiates directly with a-catenin. a- Catenin then mediates the interaction between the cadherin-catenin complex and the actin cytoskeleton. a-Catenin has been characterized as a tumor suppressor, suggesting that it may have a regulatory function in relation to its structural role. Applications: Suitable for use in Immunoblot, Immunoprecipitation, Immunohistochemistry and Immunocytochemistry. Other applications not tested. Recommended Dilution: Immunohistochemistry: Histochoice fixative is effective. Immunocytochemistry: Effective for A431 and HT1080 cell lines. Optimal dilutions to be determined by the researcher. Storage and Stability: Store product at 4°C in the dark. DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: MaxLight™490 conjugates are sensitive to light. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Note: Applications are based on unconjugated antibody.
