- Extra Details:
- The product of this gene belongs to the Serine/Threonine protein kinases family, and to the Ca(2+)/calmodulin-dependent protein kinases subfamily. Calcium signaling is crucial for several aspects of plasticity at glutamatergic synapses. This enzyme is composed of four different chains: alpha, beta, gamma, and delta. The alpha chain encoded by this gene is required for hippocampal long-term potentiation (LTP) and spatial learning. In addition to its calcium-calmodulin (CaM)-dependent activity, this protein can undergo autophosphorylation, resulting in CaM-independent activity. Two transcript variants encoding distinct isoforms have been identified for this gene. Applications: Suitable for use in ELISA and Western Blot. Other applications not tested. Recommended Dilutions: Western Blot: 0.5-2.0ug/ml Optimal dilutions to be determined by the researcher. Western Blot Analysis: 2ug/ml detected phosphorylated CaM kinase II a subunit, Mr ~50kD, in a rat brain microsomal protein preparation. Pretreating blots with lambda phosphatase (400 units/ml), prior to incubation reduced the signal strength. Treatment with lambda phosphatase had no effect on the signal strength using a non-phosphospecific CaM Kinase II antibody. A rat brain microsomal preparation was resolved by electrophoresis, transferred to nitrocellulose and incubated without lambda phosphatase or incubated with lambda phosphatase before being probed with anti-phospho-CaM Kinase II (2ug/ml). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
