Several major families of cell adhesion molecules can be distinguished. These include the cadherins and N-CAMs, which play a crucial role in cell recognition and adhesion, while integrins are implicated in the adherence of cells to the extracellular matrix. Cadherins constitute a family of proteins, the members of which are differentially expressed in the different tissues. One of the best characterized members is E-cadherin, which is prevalent in epithelial tissues. It has been shown to play a crucial role in the process of tumor cell metastasis. NCAM’s (neural cell adhesion molecules) exist in at least three forms of plasma membrane plycoproteins, which are expressed in a variety of tissues including most nerve cells. The integrins, finally, form a large family of glycosylated transmembrane proteins that act as dimers of an a- and a beta-subunit in interconnecting the cytoskeleton and the extracellular matrix. Applications: Suitable for use in Immunohistochemistry, Immunocytochemistry, and Western Blot. Other applications not tested. Recommended Dilution: Immunohistochemistry (frozen sections): 1:25-1:100, with ABC as detection reagent Western Blot: 1:50-1:500 Note: Use PBS buffer containing 0.1mM calcium chloride and 0.1mM magnesium chloride. Optimum dilutions to be determined by researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
