- The Na,K-ATPase is an integral membrane protein complex responsible for establishing the electrochemical gradients of Na+ and K+ ions across the plasma membranes of mammalian cells. The enzyme consists of an a and b subunit, both of which exist as isoforms. The a subunit is believed to be the catalytically active subunit, while the b subunit appears necessary for the assembly and transport of the sodium pump to the plasma membrane. The beta subunit is a membrane spanning glycoprotein present in all cell types. The antigenic determinant of this antibody has been mapped to the ectodomain (approximate residues 62-304) of the beta subunit. Applications: Suitable for use in Western Blot and Immunohistochemistry, Other applications not tested. Recommended Dilution: Western Blot: detects ATPase beta 1, Na, K in 20ug of rat brain microsomal preparation. A previous lot detected ATPase beta 1, Na, K using rat heart and rat kidney microsomal preparations. Rat brain microsomal preparation was resolved by electrophoresis, transferred to PVDF. Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunohistochemistry: 4% paraformaldehyde fixed tissue. Optimal dilutions to be determined by the researcher. Hybridoma: Ag8 mouse myeloma cells with immunized Balb/c splenocytes. Storage and Stability: Store product at 4°C if to be used immediately within two weeks. For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C. Aliquots are stable at -20°C for 12 months after receipt. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Note: Applications are based on unconjugated antibody.
